Dollars. The exchange rate doesn't matter in the end.
If it's a widget imported from Canada, and it's value in Canadian dollars is 50,000, then the 25% tariff would be assessed on the US value. The US value would be about $35,000 US, so the tariffs the American importer would pay would be 25% of $35,000 or $8750 US.
