Between 2003-2010, there was ONE team, ONE SINGLE TEAM, that made the final four even though they were outside the top 50 in Defensive Efficiency. That was the Dwyane Wade-powered 2003 Marquette squad, who ranked 101st.
This year, Mizzou ranked 154th. It's a major red flag for how far you should expect to advance in the tournament. In fact, the NY Times college basketball blog predicted that both Mizzou and Duke were most vulnerable to upsets based on this and a couple of other numbers:

Our offense was beautiful, but our defense was unquestionably the Achilles Heel of the 11-12 Tigers. It's very difficult to win championships with the 154th best defense in the nation. Our offense was so good, though, that it almost cancelled that out. Almost.
